Overview

L. helveticus R0052 is a probiotic strain from Institut Rosell (now Lallemand Inc., Montreal, Canada). It is often studied and sold in combination with Bifidobacterium longum R0175. It is also combined with Lacticaseibacillus rhamnosus R0011 [1]. L. helveticus stayed in the emended genus Lactobacillus (Lactobacillus sensu stricto) after the 2020 reclassification [6].

Mechanism

Surface-layer (S-layer) protein extracts from R0052 block adhesion of enterohaemorrhagic E. coli O157:H7 to cells lining the gut. This shows a direct anti-sticking mechanism (in vitro) [4]. In animal models, R0052 changes gut microbiome composition and metabolome. It relieves D-galactosamine-induced liver injury through anti-inflammatory and barrier-protective pathways (animal) [5].

Health Benefits

A randomized, double-blind, placebo-controlled trial showed that a combination of L. helveticus R0052 and L. rhamnosus R0011 reduced the duration of antibiotic-associated diarrhoea in healthy adults (RCT) [2]. An exploratory RCT in female dancers found that R0052 combined with B. longum R0175 changed endocannabinoid and inflammatory responses (RCT) [3]. A comprehensive post-market review gathered safety and efficacy data across multiple human studies for the R0052/R0011 combination. The review noted favorable tolerability (review) [1]. Evidence for psychobiotic and metabolic effects is preliminary. Reducing antibiotic-associated diarrhoea is the most robust clinical finding.

Biology

L. helveticus stayed in Lactobacillus sensu stricto. It was not moved to a new genus by Zheng et al. 2020. It is a Gram-positive, rod-shaped bacterium. It produces only lactic acid from sugar. It ferments hexoses to L-lactic acid. It has strong protein-digesting activity. It generates bioactive peptides from milk proteins. The species is linked to fermented dairy environments and the mammalian GI tract [6]. Zheng et al. (2020) kept L. helveticus within the emended genus Lactobacillus. It is closely related to L. gasseri and L. delbrueckii [6].
